JQuery is imperative.
I don't have a website that I don't use JQuery with. The best thing about it is that you can write good HTML and script it up after the fact. JQuery makes this process very easy and it's simple to learn. I'm a huge fan.

A must-have
jQuery is my first go-to when doing anything in Javascript beyond simple scripting. I've worked with Yahoo's YUI, MooTools and Prototype/Scriptaculous and felt they were all a lot harder to use with no real advantages for what I want to do. The guys at work who do heavy-duty page development are also using jQuery for major applications and feel the same way.

I pretty much add this library to every site I build these days. It makes life so much easier. And there are tons of free plugins out there too.

One of the better libraries
You need to know what you are doing if you want to use jQuery (i.e. you need to be comfortable with writing JavaScript) but if you do it will really make your life easier. While I mainly use MochiKit (another library), jQuery is the only other library I would consider using - using CSS selector syntax for querying the DOM is great and helps enforce the use of CSS in your pages. Good stuff!
